An ultra structure study on the effects of nerve growth factor and insulin-like growth factor on rat peripheralnerve regeneration

Abstract Introduction: Formation of myelin depends on maturation of the axonal cytoskeleton and mitochondria. Nerve growth factors and insulin-like growth factor can specifically play a role in their regulation. Materials&Methods: This research is an experimental study. Forty eight male rats (200-250 g) were used. After axotomy, 1 cm segment of the sciatic nerve in rats was resected, then the gap was bridged by one of the four methods (polyvinylidene fluoride or PVDF + nerve growth factor + insulin growth factor -1, autograft, sham and control). Sciatic nerve was studied by electron microscope after 4 and 12 weeks of implantation. Findings: After 4 weeks of regeneration , the mean number of unmyelinated axons in autograft and nerve growth factor groups was not significant , but after 12 weeks of implantation , the mean number of unmyelinated axons in autograft group was superior to that of the nerve growth factor group (P<0.01). After 12 weeks of regeneration, microtubular density values of normal and autograft groups were not statistically different. The cristae of axonal mitochondria in experimental groups were parallel with their long dimensions. Conclusion: These results indicate that the positive effects of nerve growth factors on formation of microtubules and mitochondria may be useful for treating peripheral nerve injuries
