Study of the Anti-Inflammatory and Analgesic Effects of Alcoholic Extract of Melilotus Officinalis L Plant in Male Mice

Introduction:  Pain is one of the most critical defense mechanisms of the body in response to tissue injury and inflammation, and its management remains a key therapeutic challenge. In recent years, medicinal plants have gained significant attention due to their bioactive compounds and lower side effects. Therefore, this study aimed to evaluate the antinociceptive and anti-inflammatory effects of the ethanolic extract of Melilotus officinalis L. in male mice.
Materials & Methods: The present experiment was performed on 30 male mice randomly divided into five groups of six animals each including negative control, positive control (treated with dexamethasone, indomethacin and morphine) and three experimental groups treated with ethanolic extract of Melilotus officinalis (prepared by 80% ethanol maceration) at doses of 100, 200 and 400 mg/kg by intraperitoneal (i.p.) injection. The anti-inflammatory and antinociceptive effects of the extract were evaluated by the xylene-induced ear edema, formalin and acetic acid-induced writhing tests. All data are presented as mean ± SEM and were analyzed with SPSS software (version 20). Differences between groups were analyzed by one-way analysis of variance (ANOVA) followed by Tukey’s post-hoc test . The threshold of significance was set to 0.05.
Results: The xylene test showed that the M. officinalis extract at 200 and 400 mg/kg significantly reduced inflammation. The dose of 400 mg/kg significantly reduced the licking/biting time in the late phase (inflammatory pain) in the formalin test. All three doses (100, 200 and 400 mg/kg) significantly reduced the number of abdominal writhes in acetic acid test compared with control group.
Conclusion: The present study results indicate that Melilotus officinalis extract has significant anti-inflammatory and antinociceptive activities. These results support the potential of this plant as a natural alternative or supplement of synthetic analgesics in traditional and modern medicine.
