تأثیر غذای پرچرب و تمرین استقامتی تحت دمای نزدیک به خنثی بر بیان ژن‌‌های پمپ کلسیم شبکۀ اندوپلاسمی (SERCA2) و گیرندۀ ریانودین (RYR2) در بافت چربی زیرپوستی موش

مقدمه: بخشی از گرمازایی غیرلرزشی در سطح بافت چربی بژ از طریق سازوکار چرخۀ بیهودۀ کلسیم سلولی به‌دست می‌آید که توسط پمپ کلسیم شبکۀ اندوپلاسمی (Sarco/Endoplasmic Reticulum ATPase2) SERCA2 و گیرندۀ ریانودین RYR2 (Ryanodine Receptor2) تنظیم می‌‌شود. در این پژوهش، اثر غذای پرچرب و تمرین استقامتی تحت شرایط دمای نزدیک به خنثی (دمایی که انسان‌‌ها زیست می‌‌کنند) بر بیان ژن‌‌های SERCA2 و RYR2 در بافت چربی زیرپوستی منطقۀ رانی موش مطالعه شد.
مواد و روش ها: 28 سَر موش سوری نر نژاد C57BL/6 به چهار گروه هفت‌تایی 1. کنترل، 2. غذای پرچرب، 3. تمرین استقامتی و 4. غذای پرچرب-تمرین تقسیم گردیدند. دمای زیست موش‌‌ها روی 26 درجۀ سانتی‌‌گراد تنظیم شد. موش‌‌های گروه غذای پرچرب به مدت 12 هفته، غذای پرچرب (45 درصد چربی) مصرف کردند. موش‌‌های گروه‌‌ تمرینی به مدت شش هفته، تحت تمرین استقامتی تداومی روی نوار گردان قرار گرفتند. موش‌‌های گروه غذای پرچرب-تمرین در کنار تغذیه با غذای پرچرب، در شش هفتۀ پایانی دورۀ مداخله، تمرین استقامتی انجام دادند. برای اندازه‌‌گیری بیان نسبی ژن‌‌هایSERCA2b,a  و RYR2 از روش Real Time–PCR استفاده شد.
یافته‌ها: آزمون تحلیل واریانس دوطرفه نشان داد بیان نسبی ژن‌‌های SERCA2a، SERCA2b و RYR2 متأثر از غذای پرچرب، تمرین استقامتی و غذای پرچرب-تمرین نشد (P˃0.05).
بحث و نتیجه‌گیری: رژیم غذای پرچرب و تمرین منظم استقامتی تحت شرایط دمای نزدیک به خنثی، تغییری در بیان تنظیم‌‌گرهای چرخۀ کلسیم در بافت چربی بژ نداشت؛ ازاین‌رو استدلال می‌‌شود که تغییرات گرمازایی غیرلرزشی ناشی از شرایط ذکرشده، احتمالاً مستقل از سازوکار چرخۀ بیهودۀ کلسیم است.
