بررسی اثربخشی کوئرستین و آنالوگ‌هایش بر آنزیم پروتئاز اصلی کووید-19 با استفاده از مطالعات داکینگ مولکولی

مقدمه: کووید-19 بیماری عفونی تنفسی حاد است که توسط ویروس SARS-CoV-2 ایجاد می شود و با توجه به شیوع سریع بیماری و علی رغم پیشرفت علمی انجام‌شده در توسعۀ واکسن، نیاز فوری به کشف داروهای ضدویروسی برای عملکرد بهتر در برابر سویه‌های جدید کروناویروس‌ها وجود دارد. در این تحقیق اثربخشی کوئرستین و آنالوگ‌هایش بر آنزیم پروتئاز اصلی کووید-19 ارزیابی شد.
مواد و روش ها: در این پژوهش توصیفی-تحلیلی، برای بررسی بیوانفورماتیکی، ساختار سه‌بعدی آنالوگ‌های کوئرستین (20 ترکیب)، داروهای استاندارد (ریتوناویر و لوپیناویر) و آنزیم پروتئاز اصلی کووید-19 به‌ترتیب از پایگاه داده های PubChem و PDB دریافت گردید. مطالعات داکینگ مولکولی ترکیبات روی پروتئاز اصلی با استفاده از نرم‌افزار MOE.2014 انجام شد؛ سپس خصوصیات فیزیکوشیمیایی و فعالیت بیولوژی ترکیبات با استفاده از نرم‌افزارهای Swiss ADME، PASS وSwiss Target Prediction پیش‌بینی گردید.
یافته‌ها: یافته های مطالعۀ حاضر نشان داد که مهم ترین پیوند های درگیر در اتصال دارو با گیرنده، پیوند هیدروژنی، برهمکنش های هیدروفوب و π-π هستند. بهترین نتایج داکینگ مربوط به ترکیبات Baicalein، Genistein، Naringenin و Quercetin با داشتن انرژی اتصال قوی (83/12- تا 54/13- کیلوکالری بر مول)، در مقایسه با داروهای ریتوناویر و لوپیناویر است. این ترکیبات برای اتصال با آمینواسیدهای کاتالیتیکی His41 و Cys145 و دیگر آمینواسیدهای کلیدی جایگاه فعال آنزیم پروتئاز اصلی تمایل بیشتری دارند.
بحث و نتیجه‌گیری: بر اساس نتایج به‌دست‌آمده از مطالعات بیوانفورماتیکی، آنالوگ‌های کوئرستین به‌سبب قرارگیری مناسب در جایگاه فعال آنزیم پروتئاز اصلی کووید-19، مهار مؤثرتری را نسبت به داروهای شیمیایی استاندارد ایجاد می کنند و می توانند کاندید های مناسبی برای بررسی های in vitro و in vivo باشند.
