بررسی فعالیت آنتی‌اکسیدانی و تعیین محتوای فنولی و فلاونوئیدی گیاه دارویی Tussilago farfara L. در طبقات ارتفاعی مختلف و مراحل فنولوژی

مقدمه: گیاه علفی چندسالۀ پای خر (.Tussilago farfara L) از خانوادۀ Asteraceae است که برگ‌ها و گل‌های آن خاصیت دارویی دارد و از مواد مؤثر آن برای معالجۀ سرفه‌های خشک و درمان التهاب‌های دهان، گلو و مجاری تنفسی استفاده می‌شود؛ بنابراین، هدف از این مطالعه بررسی برخی ویژگی های فیتوشیمیایی گیاه پای خر (Tussilago farfara L.) و مقایسۀ تغییرات آن در طبقات مختلف ارتفاعی و مراحل فنولوژی است.
مواد و روش ها: برای بررسی روند تغییرات میزان مادۀ مؤثر گیاه موردبررسی در گرادیان ارتفاعی، با توجه به حضور و فراوانی گونه، طبقات ارتفاعی 1100 تا 2600 متری در استان گلستان انتخاب شد. به‌منظور نمونه‌برداری از گیاه در مراحل فنولوژی (پیش از گلدهی، زمان گلدهی و پس از گلدهی) در هر طبقۀ ارتفاعی، به‌طور تصادفی از گیاه در هریک از پنج طبقۀ ارتفاعی، نمونه­ها جمع‌آوری و به آزمایشگاه منتقل گردید. در پژوهش حاضر محتوای فنول کل با استفاده از روش فولین سیوکالتو، محتوای فلاونوئید با روش آلومینیوم کلرید و همچنین بررسی فعالیت آنتی‌اکسیدانی عصارۀ متانولی تهیه‌شده با استفاده از معرف DPPH صورت گرفت.
یافته‌ها: نتایج نشان داد، بیشترین مقادیر محتوای فنول کل، فلاونوئید و فعالیت آنتی‌اکسیدانی در ارتفاعات بالاتر از 2200 متر است و اختلاف معنی‌داری میان طبقات ارتفاعی به لحاظ ویژگی‌های فیتوشیمیایی وجود داشت. یافته‌های تحقیق در بررسی و مقایسۀ خصوصیات فیتوشیمیایی گیاه در میان مراحل مختلف فنولوژی نشان داد، بالاترین مقدار محتوای فنول کل و فلاونوئید در مرحلۀ فنولوژی گلدهی و بالاترین میزان فعالیت آنتی‌اکسیدانی برگ گیاه در مرحلۀ رویشی با مقدار 08/42 درصد مشاهده شد.
بحث و نتیجه‌گیری: نتایج این مطالعه اطلاعات ارزشمندی در تعیین دامنۀ ارتفاعی و مرحلۀ فنولوژی مناسب در برداشت برگ این گیاه، به‌منظور دستیابی به بالاترین مقادیر محتوای فنول، فلاونوئید و فعالیت آنتی‌اکسیدانی، برای دسترسی فعالان در زمینۀ گیاهان را دارویی فراهم آورد.
