آنالیز بیوانفورماتیکی تأثیر Linalool تشنه‌داری(Scrophularia striata) بر پروتئین VacA هلیکوباکتر پیلوری

مقدمه: برخی گیاهان ازجمله تشنه­داری، به‌طور سنتی در میان مردم زاگرس‌نشین، به‌عنوان التیام‌بخش عفونت و زخم استفاده می­شده است. در تحقیقات پیشین، اثر التیام‌بخشی عصارۀ آبی و هیدروالکلی آن بر استرس اکسیداتیو و درمان زخم معدۀ ناشی از اتانول در موش صحرایی تائید شده است. بر اساس این، هدف از تحقیق حاضر، آنالیز بیوانفورماتیکی تأثیر Linalool (یکی از بیشترین اجزای تشنه­داری) بر مهار پروتئین VacA هلیکوباکتر پیلوری است.
مواد و روش‌ها: توالی­های DNA و پروتئینی Linalool گیاه آرابیدوپسیس و توالی ژن VacA هلیکوباکتر پیلوری از سراچۀ NCBI تهیه شد. میزان هم‌ردیفی توالی‌های DNA و پروتئینی Linalool و VacA توسط نرم‌افزار Clustalw vol.2  بررسی و موتیف­های حفاظت‌شده با استفاده از برنامه MEME جستجو گردید. درخت فیلوژنی به‌وسیلۀ نرم­افزار MEGA vol.5 ترسیم شد. برای تعیین ساختار سوم و همچنین داکینگ مولکولی بر اساس همولوژی، توسط نرم‌افزار Swiss-Model و ESYPred3D و همچنین Galaxy Web صورت گرفت.
یافته‌های پژوهش: پروتئین‌های لینالول گزارش‌شده در سراچۀ NCBI، در بیشتر گیاهان، سه موتیف حفاظت‌شده (در محدودۀ 100 تا 300 اسید آمینه) دارد. پروتئین VacA هر سه نوع دومین سیتوپلاسمی، غشایی و کینازی را داراست. بهترین الگوی داکینگ مولکولی بر اساس بیشترین اثر متقابل میان پروتئین لینالول و پروتئین VacA (0/21) و بیشترین دقت (133/0) انتخاب شد. نتایج نشان داد که در ساختار هر دو پروتئین لینالول و VacA، گروه‌های گوناگونی ازجمله گروه‌های آلیفاتیک (چربی)، قطبی و آب‌دوست داشت. حضور چربی و آب‌دوست بودن در پروتئین VacA، به آن این قابلیت را می‌دهد که بتواند در صورت تجزیه نشدن، توسط سلولی‌های لنفوسیت، به‌راحتی از غشای سلول‌ها عبور کند و سبب بیماری‌زایی شود. از سویی، حضور همین گروه‌ها در لینالول نیز عاملی است که توانسته است پروتئین VacA را مهار کند.
بحث و نتیجه‌گیری: آنالیز بیوانفورماتیکی نشان داد که لینالول سبب از میان بردن آثار هلیکوباکتر پیلوری می­شود؛ همچنین نتایج پژوهش حاضر می‌تواند به‌عنوان پایه‌ای برای تحقیقات آینده، برای ارزیابی تأثیر تک­تک مواد مؤثر تشنه­داری به‌ویژه لینالول، بر باکتری هلیکوباکتر پیلوری استفاده شوند.
