@article{ author = {Naserifar, R and Ghaffarifar, F and DalimiAsl, A and Sharifi, Z}, title = {Cloning of Toxoplasma Gondii Granular Antigen 5GRA)5)}, abstract ={Abstract Introduction: Toxoplasmosis is a one of the most world-wide spread zoonosis representing a very serious clinical and veterinary problem. Toxoplasma gondii is an intracellular parasite that causes severe neurologic and ocular disease in immune compromised and congenitally infected individuals. The heavy incidence and severe or lethal damages of toxoplasmosis clearly indicate the need for the development of a more effective vaccine human vaccines are not available and current anti-toxoplasma treatment is disappointing. Immunization with plasmid DNA, a relatively novel technique, is a promising vaccination technique. To improve the immune response by DNA vaccination various is one of the key for the success of the vaccine in the field. One of the most efficient ways to control this disease is immunization. However, so far, there is no effective vaccine available against this pathogen. An important source of human contamination with T. gondii is the consumption of raw or undercooked meat products. Toxoplasma gondii are widely prevalent in humans and other animals which can cause severe or lethal toxoplasmosis. So, the development of a more effective vaccine is needed urgently .Therefore, we prepare gra5 plasmid to use as a vaccine. Materials & Methods: In this study, GRA5was cloned in pTZ57R afterwards, it was transformed into TOP10 strain of E.coli Bacteria. The recombined plasmid extracted from E.coli bacteria and amplified through PCR technique. Besides, pcDNA3 Plasmid for receiving and cloning of GRA5 segment was digested by Hind3 & EcoRI enzymes. GRA5 was sub-cloned into pcDNA3 and the reaction ligation product was transformed for the above bacteria. The bacteria grew in LB culture with ampicillin. Recombined pcGRA5 plasmids were purified from E.coli by Plasmid extraction kit. Finally, recombinant plasmid using cell culture method was expressed in Cho cell. Findings: The accuracy of the results was confirmed by using restriction enzymes and PCR methods. GRA5 was cloned into expression eukaryotic plasmid pcDNA3. After sequencing pcGRA5 plasmid for cell expression Western blot method was used. Discussion & Conclusion: The results showed that cloning and transformation of fragment GRA5 in pcDNA3 was done properly.}, Keywords = {Immunization, GRA5, toxoplasma gondaii, cell expressio n, DNA vaccine}, volume = {19}, Number = {3}, pages = {1-12}, publisher = {Ilam University of Medical Sciences}, url = {http://sjimu.medilam.ac.ir/article-1-508-en.html}, eprint = {http://sjimu.medilam.ac.ir/article-1-508-en.pdf}, journal = {Journal of Ilam University of Medical Sciences}, issn = {1563-4728}, eissn = {2588-3135}, year = {2011} } @article{ author = {Shakib, P and Sadeghifard, N and Zolfaghary, M and Gafouryan, S and Maleki, A and Mohebi, R and Ranjbar, R}, title = {Frequency of Ompk35 And Ompk36 in Strains of}, abstract ={: Klebsiella pneumonia is a major nosocomial pathogen causing pneumonia, urinary tract infections, and bacteremia, particularly in immune-compromised patients. The aim of our study was expression of OMPK36 and OMPK35 in ESBLs production and non-ESBLs K.pneumoniae. Materials & Methods: Clinical isolates of K.pneumoniae were identified. ESBLs were assayed by Phenotypic (screening and confirming) and genotypic (PCR) methods. OMPK35 and OMPK36 were tested by polymerase chain reaction and SDS- PAGE. Findings: Our findings showed that 42.3% of K.pneumoniae were ESBLs positive. Twenty-two K.pneumoniae producing ESBLS and Twenty-two non-ESBLs K.pneumoniae were selected for porines detection. We found out that 54.54% and 72.72% of K.pneumoniae producing ESBLS were positive for Ompk35 and Ompk36 respectively, while our findings in non-ESBLs K.pneumoniae showed that 95.94% and 100% of isolates were positive for Ompk35 and Ompk36, respectively. Discussion & Conclusion: Many of clinical isolates of non- ESBL K. pneumoniae had expressed both Ompk35 and Ompk36 porins, while our results in ESBLs positive K.pneumoniae had only showed expression of Ompk36. The aim of this study was to investigate the acute effects of eccentric and concentric exercise on oxidant and antioxidant factors in active young women. Materials & Methods: Twenty four voluntary female students participated in the study, who were randomly assigned to three groups: control (no training), eccentric training group (ellestad test with reverse slope), and concentric training group (ellestad test with straight slope). Blood samples were collected 1h before and immediately after exercise for assessment of plasma total antioxidant capacity (TAC), glutathione (GSH), and malondialdehyde (MDA). The SPSS software Ver.13 was used, while one-way variance was applied to determine dependent variables. T-test analysis was performed on values from blood samples to obtain differences between pre-and post training values. Findings: The results showed that after eccentric and concentric exercise, there were a significant increase in plasma TAC, GSH, MDA levels compared with that of what before the exercise, (p<0.05). Factors that can affect hemodynamic and O2sat include: type of anesthetic, duration of surgery, transportation, age, previous underlying disease, …. There are poorly documented studies about such changes in children. So, the goal of this study was to evaluate the hemodynamic and O2sat variabilities among children at recovery room. Materials & Methods: In this prospective-discriptive study, 120 children aged 1-12 years undergoing minor surgery with general anesthesia were investigated. Systolic, diastolic and mean arterial pressure and O2sat were measured before the anesthesia and surgery, immediately before transfer to recovery and after arrival at recovery room. Data was analyzed by SPSS software. Findings: Mean systolic, diastolic, mean arterial blood pressure and O2sat in recovery room decreased and pulse rate increased significantly compared with the other times, (P<0.05). Discussion & Conclusion: Hypotension can be due to hypoxemia, decreasing in venous return and cardiac output. Tachycardia may be the result of compensatory response to hypotension, pain, hypoxia, and the stress due to extubation. Hypoxia may indicate upper airway obstruction, atelectasia and anesthetic depression.
