fa مقایسۀ نیمرخ ژنتیکی چندشکلی تک نوکلئوتیدی rs4646994 ژن ACE در مردان نخبۀ وزنه بردار با غیرورزشکاران Comparison of the Genetic Profile of rs4646994 Single Nucleotide Polymorphism of the ACE Gene in the Elite Male Weightlifters and Non-athletes تربیت بدنی Physical Education پژوهشي Research مقدمه: ژن ACE آنزیم مبدل آنژیوتانسین (ACE) را کدگذاری می کند که با وجود آلل I یا نبودِ آلل D یک قطعه 287 جفت بازی مشخص می شود. آلل D با فعالیت ACE بالاتر و افزایش سطح آنژیوتانسین II و عملکردهای ورزشی مبتنی بر قدرت همراه است؛ بنابراین، هدف مطالعۀ حاضر مقایسۀ نیمرخ ژنتیکی چندشکلی تک نوکلئوتیدی rs4646994 ژن ACE در مردان نخبۀ وزنه بردار با غیرورزشکاران بود. مواد و روش ها: 30 مرد نخبۀ وزنه بردار (قهرمانان لیگ، کشور، تیم ملی، قهرمانان آسیا، جهان و المپیک) مقیم استان اصفهان، به همراه 43 داوطلب غیرورزشکار سالم که از لحاظ سن، قد و وزن مشابه گروه نخبه بودند، آزمودنی های بررسی را تشکیل دادند. با تکمیل فرم رضایت‌نامه، نمونه های بزاق از شرکت کنندگان جمع آوری شد و پس از مراحل استخراج DNA، تعیین ژنوتیپ های ACE با استفاده از تکنیک های PCR_RLFP و الکتروفورز صورت گرفت. تجزیه‌وتحلیل داده ها با استفاده از روش های آماری، t مستقل، کای مربع و رگرسیون لجستیک از طریق نرم افزار SPSS vol.20 انجام گردید. یافته‌ها: نتایج نشان داد شیوع آلل D به‌طور معنی داری در وزنه برداران نخبه و غیرورزشکاران، بالاتر از آلل I در گروه ای خود بود (0.05<P)؛ همچنین ژنوتیپ D/I هم در جمعیت مطالعه‌شده (7/50 درصد) و هم در دو گروه وزنه برداران (7/56 درصد) و غیرورزشکاران (5/46 درصد) شایع ترین بود؛ اما اختلاف معنی داری در توزیع ژنوتیپینگ ACE I/D در وزنه برداران نخبه با غیرورزشکاران وجود نداشت (0.05<P،0.31 =χ2). بحث و نتیجه‌گیری: مطالعه نشان می دهد ارتباط مثبتی میان وزنه برداران نخبۀ ایرانی و آلل ACE D وجود دارد که احتمالاً آن را به عنوان یکی از عوامل ژنتیکی مؤثر در ایجاد یک فنوتیپ قدرت وزنه‌برداران ایرانی متمایز کرده است. Introduction: The ACE gene encodes the angiotensin-converting enzyme (ACE), which is characterized by the presence of allele I or the absence of allele D of a 287-bp fragment. The D allele is associated with higher ACE activity and increased angiotensin II levels and performance in strength-oriented exercise tasks. This study aimed to compare the genetic profile of rs4646994 single nucleotide polymorphism of the ACE gene in the elite male weightlifters and non-athletes. Material & Methods: In total, 30 elite male weightlifters (league, country, national team, Asian, world, and Olympic champions) living in Isfahan province along with 43 healthy non-athlete volunteers who were matched in age, height, and weight to the elite group formed the subjects of the study. After completing the consent form, saliva samples were collected from the participants, and after DNA extraction, ACE genotypes were determined using PCR-RLFP and electrophoresis techniques. Data analysis was performed using independent t-test, chi-square, and logistic regression in SPSS software (version 20). (Ethic code: 7993) Findings: The results showed that the prevalence of D allele in elite weightlifters and non-athletes was significantly higher than allele I in their groups (P<0.05). Moreover, in the genotyping distribution, D/I was most common both in the study population (7.50%), as well as weightlifters (7.56%) and non-athletes (5.46%). However, there was no significant difference in the elite weightlifters and non-athletes regarding the distribution of D/I ACE genotyping (χ2=0/31).
