fa بررسی واریانت ژنتیکی 45bp ins/del در ژن UCP2 به‌عنوان یک فاکتور خطر برای اختلال دوقطبی Evaluation of 45bp Ins/Del Genetic Variant in UCP-2 Gene as a Risk factor for Bipolar Disorder بیولوژی biology پژوهشي Research مقدمه: اختلال دوقطبی نوعی اختلال دوفازی از در دسترس بودن انرژی است که نشان‌دهندۀ افزایش تنفس میتوکندریایی در فاز شیدایی، در مقایسه با کاهش عملکرد میتوکندری در فاز افسردگی بیماری است. نقص در عملکرد میتوکندری، به افزایش بیش‌ازحد گونه‌‌های فعال اکسیژن (ROS) منجر می‌‌شود. پروتئین‌‌های UCP-2 به‌عنوان تنظیم‌کننده‌های ROS در سلول عمل می‌‌کنند. هدف از این مطالعه، بررسی همراهی واریانت ژنتیکی 45bp ins/del در ژن UCP-2 با استعداد ابتلا به اختلال دوقطبی است. مواد و روش‌ها: این مطالعه مورد-شاهدی، بر روی 205 فرد مبتلا به اختلال دوقطبی و 200 فرد سالم به‌عنوان گروه کنترل انجام گرفت. پس از استخراج DNA از نمونه‌‌های خون محیطی، تعیین ژنوتیپ به‌‌وسیلۀ روش PCR انجام شد؛ همچنین آنالیز رگرسیون لجستیک برای مقایسۀ فراوانی آللی و ژنوتیپ‌‌ها میان گروه کنترل و بیمار استفاده گردید. یافته‌ های پژوهش: ارتباط معنا‌‌داری میان ژنوتیپ‌‌های ID(P˂0.001، 95% CI=0.27-0.64، OR=0.42) و II (P=0.006، 95% CI=0.20-0.77، OR=0.39) و استعداد ابتلا به اختلال دوقطبی وجود داشت. آنالیز داده‌ها نشان داد که آلل I اثر حفاظتی بر ابتلا به اختلال دوقطبی دارد (P˂0.001، 95% CI=0.42-0.75، OR=0.56). بحث و نتیجه‌گیری: در این مطالعه، برای نخستین بار، همراهی میان واریانت ژنتیکی UCP-2 45bp ins/del و اختلال دوقطبی بررسی شد. نتایج نشان داد که واریانت ژنتیکی 45bp ins/del در ژن UCP-2 با اختلال دوقطبی همراهی دارد. مطالعات بیشتر در جمعیت‌‌های نژادی مختلف، برای تأیید نتیجۀ مطالعه حاضر لازم است. Introduction: Bipolar disorder is a type biphasic disorder of energy availability that indicates increased mitochondrial respiration in episodes of mania, compared to decreased mitochondrial function in the depressive phase of the illness. Mitochondrial dysfunction may lead to an excessive increase in reactive oxygen species (ROS). The UCP-2 proteins act as regulators of ROS in the cell. This study aimed to investigate the association of UCP-2 45bp ins/del genetic variant with susceptibility to bipolar disorder.   Materials & Methods: This case-control study was performed on 205 subjects with bipolar disorder and 200 healthy subjects as a control group. After extraction of DNA from peripheral blood samples, genotype determination was performed using the PCR method. Moreover, the logistic regression analysis was used for the comparison of genotypes and allele frequencies between case and control groups. Ethics code: IR.SUMS.REC.1396.S435   Findings: There was a significant association between ID (OR=0.42; 95% CI= 0.27-0.64; P<0.001) and II (OR=0.39; 95% CI=0.20-0.77; P=0.006) genotypes with bipolar disorder. Data analysis showed I allele had a protective effect on susceptibility to bipolar disorder (OR=0.56; 95% CI=0.42-0.75; P<0.001).   Discussions & Conclusions: This study investigated, for the first time, the association between UCP-2 45bp ins/del genetic variant and bipolar disorder.
