اثر سیلیمارین بر مسیر میتوفاژی در سلول‌های سرطانی کولون انسانی HT-29

مقدمه: آثار ضدسرطانی سیلیمارین در مطالعات متعددی گزارش‌شده است. با وجود این، سازوکار اثر آن در سلول‌های سرطانی کولون به‌درستی شناخته‌نشده است. در این مطالعه اثر سیلیمارین بر مسیر میتوفاژی و ایجاد گونه­های آزاد اکسیژن (ROS) در سلول‌های سرطانی کولون انسانی (HT-29) بررسی‌شده است.
مواد و روش ها: سلول‌های سرطانی HT-29 و سلول‌های غیرسرطانی NIH-3T3 به مدت 48 ساعت در معرض غلظت‌های 0 (کنترل)، 25، 50 و 100 میکروگرم بر میلی‌لیتر سیلیمارین قرار گرفتند. درصد زنده­مانی سلول‌ها، سطح ROS، پتانسیل غشای میتوکندری (MMP) و بیان ژن‌های مهم مسیر میتوفاژی شامل Pink1 (PTEN-induced putative kinase protein 1)  و Parkin برای ارزیابی آثار سیلیمارین بررسی گردید.
یافته‌ها: سیلیمارین به‌صورت وابسته به غلظت، درصد زنده­مانی سلول‌های HT-29 را به‌طور معنی‌داری کاهش داد (P<0.05)؛ همچنین سیلیمارین به‌صورت وابسته به غلظت، سبب کاهش معنی‌داری MMP شد، درحالی‌که تشکیل ROS را در سلول‌های HT-29 به‌طور معنی‌داری افزایش داد (P<0.05). علاوه بر این، سیلیمارین بیان ژن‌های Pink1 و Parkin را به‌طور معنی‌داری و به‌صورت وابسته به غلظت افزایش داد (P<0.05). غلظت 100 میکروگرم بر میلی‌لیتر سیلیمارین بیشترین تأثیر را در کاهش درصد زنده­مانی و MMP و همچنین افزایش سطح ROS و بیان ژن‌های مرتبط با میتوفاژی داشت (P<0.001). سیلیمارین در غلظت‌های مختلف تغییر معنی‌داری در درصد زنده­مانی، سطح ROS و MMP در سلول‌های غیرسرطانی NIH-3T3 ایجاد نکرد.
بحث و نتیجه‌گیری: داده­های این مطالعه نشان می­دهند که سیلیمارین احتمالاً با کاهش MMP، سبب افزایش سطح ROS و درنتیجه، القای میتوفاژی از طریق مسیر سیگنالینگ Pink1 / Parkin در سلول‌های HT-29 می­شود.
