پیشگویی بیوانفورماتیکی ژن‌های غیرکدکنندۀ مرتبط با ژن‌های مسیر تمایز اکتودرمی FGF8، NOG و BMP4 موشی و ترسیم شبکۀ ارتباطی آن‌ها

مقدمه: lncRNAها و miRNAها دسته‌ای از RNAهای غیرکدکننده هستند که نقش و عملکردهای مهمی در تنظیم بیان ژن‌ها و فرایندهای اصلی بیولوژیکی ازجمله تکثیر سلولی، آپوپتوز و تمایز دارند. lncRNAها می‌توانند به‌طور بالقوه روی miRNAها، در جهت همسو و یا معکوس بر فعالیتشان تأثیر بگذارند تا از این طریق، نقش تنظیم‌کنندگی آن‌ها را تعدیل کنند. در این مطالعه، شبکه‌های ژنی mRNA-miRNA-lncRNA با استفاده از برنامه‌های آنلاین برای سه مارکر مسیر اکتودرمی (BMP4,NOG,FGF8) در سلول‌های بنیادی جنینی موشی ترسیم شد.
مواد و روش ها: این مطالعه از نوع تئوریکال بیوانفورماتیکی است و micRNAهای هدف ژن‌های BMP4، NOG و FGF8 توسط پایگاه‌های داده‌ای MirWalk و TARGETSCAN استخراج و بررسی گردید تا درنهایت، بتوان miRNAهای مشترک این ۳ ژن را تهیه کرد؛ سپس از پایگاه داده‌ای DIANA-Tools، lncRNAهای هدف برای miRNAهای مشترک به‌دست آمد.
یافته‌ها: ژن‌های mmu-miR-92a-2-5p، mmu-miR-129b-5p، mmu-miR-130b-5p، mmu-miR-692، mmu-miR-7009-3P، mmu-miR-7116-3p و mmu-miR-7689-3p ممکن است بر فعالیت lncRNA‌های Kcnq1ot1، Gm26812، Gm4117، Gm11837.4930423MO2Rik، Malat1، Gm12594، Gm3414.5830444B04Rik، Gm2464 و NEAT1 اثر بگذارند.
بحث و نتیجه‌گیری: با توجه به ارتباط متقابل lncRNA، miRNA و mRNA، مطالعۀ ما یک نقش جدید از lncRNAها را برای تحقیقات آینده و مطالعات تجربی دربارۀ مسیر تمایز اکتودرمی و سازوکار‌های مولکولی فراهم می‌کند.
