بهینه‌سازی کشت سلول‌های دندریتیک نابالغ از سلول‌های مغز استخوان موش در شرایط عاری از اندوتوکسین

مقدمه: سلول‌های دندریتیک در مطالعات پایه و همچنین تحقیقات دربارۀ واکسن بسیار اهمیت دارند؛ اما جداسازی و کشت این سلول‌ها به علت تعداد اندک در بافت‌ها، با چالش‌هایی روبروست. با توجه به اینکه روش استانداردی وجود ندارد، در این پژوهش، به برخی از عوامل مؤثر در افزایش بازدهی جداسازی سلول‌های دندریتیک از مغز استخوان موش BALB/c پرداخته‌شده است.
مواد و روش‌ها: پس از جداسازی سلول‌های مغز استخوان از فمور و تیبیا و هم‌جواری با سایتوکاین GM-CSF، در روز سوم، نیمی از سلول‌ها به پلیت جدید انتقال داده و در روز پنجم، از اولیگونوکلئوتید CpG برای تحریک سلول‌ها استفاده شد. در روزهای پنجم و هفتم، بقای سلول‌ها و همچنین شاخص‌های فنوتیپی CD11C، MHC-II و CD86 با فلوسایتومتری بررسی گردید.
یافته‌های پژوهش: در این مطالعه مشخص شد که حضور اندوتوکسین در مراحل کشت سلول‌های نابالغ، به‌طور معنی‌دار (P<0.05) سبب کاهش بازیابی سلول‌های دندریتیک می‌شود. علاوه بر این، تعداد کمتر از یک میلیون سلول در پلیت‌های غیرچسبنده سبب بقای بیشتر و بازدهی بالا می‌گردد. انتقال سلول‌ها در روز سوم به پلیت جدید هم به‌عنوان راهکاری برای جلوگیری از بلوغ زودرس سلول‌های دندریتیک و مرگ ناشی از آن معرفی شد. پس از دست‌یابی به سلول‌های نابالغ در مجاورت GM-CSF، می‌توان برای القای مسیر Th1 از اولیگونوکلئوتید CpG در روز هفتم استفاده کرد. اختلاف معنا‌دار (P<0.05) با کنترل نشان داد که CpG می‌تواند به‌عنوان محرک در القای بلوغ پس از تیمار با GM-CSF به‌کار گرفته شود.
بحث و نتیجه­گیری: بهینه‌سازی کشت سلول‌های دندریتیک به لحاظ شرایط مختلف کشت، نقش مهمی در بازدهی بازیابی این سلول‌ها دارد.
